Near-infrared fluorescent catheter or two-dimensional molecular imaging of coronary high-risk plaque

At the 2010 World Molecular Imaging Congress (WMIC), a collaborative study between the United States and Germany showed that near-infrared fluorescence (NIRF) catheters can perform two-dimensional molecular imaging of plaque inflammation with high dynamic range, which can be converted into clinical targets. An effective means of monitoring high-risk plaques in coronary arteries.
At present, it is still a challenge to achieve effective monitoring of high-risk coronary plaques before rupture. One of the reasons is that existing detection methods are difficult to meet the requirements for resolution and sensitivity. This study developed this NIRF catheter system for two-dimensional intravascular molecular imaging of inflammatory plaques.
The NIRF catheter system is based on the excitation light (750 nm) emitted by the fiber to the tissue and then collects the fluorescent emission. The head of the fiber has a 45 degree angled bevel that reflects the guided laser light to the vessel wall and excites the fluorescent dye of interest on the vessel wall. The dichroic mirror and filter selectively transmit fluorescence to a photomultiplier tube (PMT). The voltage generated by the PMT is used for two-dimensional fluorescent probe imaging of blood vessels.
The researchers said the system has been tested in the abdominal aorta of rabbits with hyperlipidemia and balloon injury models. Animal models were injected with a cysteine ​​proteolytic enzyme-activated NIRF probe to target inflammatory cysteine ​​proteolytic enzymes within the plaque (excitation/emission of light at 750/770 nm). The aorta was imaged using a two-dimensional intravascular imaging catheter and anatomical image calibration was performed by intravascular ultrasound (IVUS). Ex vivo fluorescence imaging (FRI) of the aorta is then continued. The results showed consistent data on the FRI of the in vivo and isolated aorta, suggesting the high resolution and sensitivity of the NIRF catheter system, as well as good clinical application prospects.
